Objective To investigate the anti-exercise fatigue effects and mechanism of Kaempferol(Kae).Methods The rats were randomly divided into control group,model group and experimental-L,-M,-H groups with 12 cases per group.Experimental-L,-M,-H groups were given 50,100 and 200 mg·kg-1 Kaempferol,respectively.Model and control groups were given equal amounts of 5%carboxymethylcellulose sodium by intragastric administration.Five groups were treated for 4 weeks with once a day.Rats in model group and experimental-L,-M,-H groups were given exhaustive swimming exercise at the 2nd-4th week.The contents of liver glycogen,serum lactate(LA)levels and superoxide dismutase(SOD)levels of gastrocnemius were measured according to the kit.The expression levels of phosphorylated-AMP dependent protein kinase α(p-AMPKα)in gastrocnemius was detected by Western blot.Results The contents of liver glycogen in control group,model group and experimental-M,-H groups were(11.22±0.71),(2.96±0.20),(5.20±0.64)and(6.06±0.42)mg·g-1;the serum LA levels were(0.31±0.03),(0.90±0.03),(0.50±0.03)and(0.40±0.03)mg·L-1;the SOD levels in gastrocnemius muscle were(20.12±0.80),(30.02±2.70),(40.04±2.31)and(45.56±1.77)U·mg prot-1;the relative expression levels of p-AMPKα/AMPKα were 0.60±0.03,0.19±0.02,0.45±0.11 and 0.49±0.05;the relative expression levels of PGC-1α were 0.80±0.03,0.10±0.01,0.49±0.10 and 0.53±0.11,respectively.Compared with the model group,the above indexes of control group and experimental-M,-H groups were statistical significance(all P<0.05).Conclusion Kaempferol can improve the anti-fatigue ability of exercise-induced fatigue rats,improve energy metabolism,reduce accumulation of harmful metabolites,increase antioxidant activity,and activate AMPK/PGC-1α signaling pathway.
