Kompetitive Allele Specific PCR(KASP)Assays for Quality Functional Genes in Yunhan Series Wheat Cultivars
In order to understand the composition of functional genes of the quality,high-throughput kompeti-tive allele specific PCR(KASP)assay related to high-molecular weight glutenin subunits,grain hardness and flour color was used to detect 24 Yunhan series wheat cultivars(lines).The results indicated that 1RS/1BL and 1RS/1AL translocation line was not detected in all tested cultivars(lines).Mutant Wx-B1b of waxy gene Wx-B1 accounted for 8.33%.Elite alleles Ppo-A1b of oxidase gene for flour color accounted for 12.5%,Ppo-D1a accounted for 16.67%,Lox-B1a accounted for 91.67%and TaPod-A1b accounted for 4.17%.The mutant Pinb-D1b of the grain hardness gene Pinb-D1 accounted for 91.67%,the mutant Pinb-B2b of Pinb-B2 accounted for 25%,and the mutant Pina-D1b of Pina-D1 was not detected.The proportion of wheat materials with gene of favorable high molecular weight glutenin subunit at Glu-A1 site was 83.33%,the proportion of wheat materials with gene of favora-ble high molecular weight glutenin subunit at Glu-B1 site was 4.17%,and the proportion of wheat materials with gene of favorable high molecular weight glutenin subunit at Glu-D1 site was 33.33%.This study clarified the com-position of quality functional genes of Yunhan series wheat varieties(lines),and provided reference for quality im-provement of wheat varieties in Shanxi province.
