Cloning and Analysis of Granule-Bound Starch Synthase Gene DaGBSS in Dioscorea alata
Granule-bound starch synthase(GBSS)is a key enzyme that can determine the biosynthesis of amy-lose.A DaGBSS gene with complete ORF was obtained by searching transcription group of Dioscorea alata and molec-ular cloning.The cDNA fragment was 1 845 bp in length and encoded 614 amino acids.Bioinformatics analysis indi-cated that DaGBSS protein was an acidic stable protein,containing two functional domains of starch synthase catalysis and glycosyltransferase.The results of multi-sequence alignment indicated that DaGBSS protein of D.alata had high homology with the selected species and contained typical functional domains.The results of biological evolution analysis indicated that DaGBSS protein was more closely related to the species evolution of Dioscorea cayenensis sub-sp.rotundata.The tuber of D.alata harvested for 15 days were treated,the results of qRT-PCR and index analysis indicated that the expression of DaGBSS gene in the root of D.alata showed obvious temporal and spatial differences,and the expression of DaGBSS gene,the content of GBSS and amylose were the highest in the part farthest from the surface.Through analysis of the physiological and key regulatory factors in the early stage of D.alata tuber starch synthesis,the study provides references for exploring the regulation of D.alata starch synthesis,and also provides important genetic resources for molecular breeding of D.alata.
