摘要
建立不同棉籽形态快速无损棉籽蛋白质含量检测的方法,对于合理有效地利用棉籽资源和开展棉籽营养品质遗传改良等研究具有关键作用.研究通过凯氏定氮法对187份棉花品种进行棉籽蛋白质含量化学值测定,利用近红外光谱分析仪分别采集3种不同棉籽形态毛籽、光籽和种仁的光谱特征信息,对原始光谱进行不同数学处理和散射处理,结合改良偏最小二乘法建立了毛籽、光籽和棉仁3种棉籽蛋白质含量快速无损检测的近红外光谱模型.模型的定标相关系数分别为0.957、0.971、0.989,并通过外部验证和统计分析,结果表明3种模型都具有良好的准确性和稳定性,能够针对不同棉籽形态进行准确的蛋白质含量测定.实现了棉花种子蛋白质含量的快速无损低消耗检测,为棉籽综合利用以及棉籽高品质育种提供参考.
Abstract
The establishment of a rapid and non-destructive method for the detecting cottonseed protein content in different morphological types plays a key role in the effective utilization of cottonseeds resources and the genetic im-provement of cottonseed nutritional quality.In this study,the reference value of cottonseed protein content of 187 va-rieties were measured by the Kjeldahl method,and the spectral characteristics of the three kinds of cottonseed of fuzzy cottonseed,shell-intact cottonseed and cottonseed kernel were collected by near infrared spectroscopy(NIRS).The original spectra were subjected to various mathematical and scattering treatments.By combining modified partial least squares(MPLS)regression,NIRS models for the rapid and non-destructive detection of protein content in fuzzy cottonseed,shell-intact cottonseed and cottonseed kernel were established.The regression squared(RSQ)of the three models were 0.957,0.971 and 0.989,respectively.External validation and statistical analysis confirmed that three models had excellent accuracy and stability.The results indicated that the NIRS detection models can be used to detect the protein content of different kinds of cottonseeds samples accurately.This achievement enabled rapid,non-destructive and low-consumption detection of protein content in cottonseeds,providing references for the com-prehensive utilization and high-quality breeding of cottonseeds.
