山桐子粕多酚提取工艺优化及抗氧化活性的研究

以山桐子粕为研究对象,选取乙醇体积分数、料液比、超声温度、超声时间进行单因素试验,评价各因素对山桐子粕多酚提取率的影响,采用响应面法优化山桐子粕多酚提取工艺.通过真空冷冻干燥得到一种具有一定抗氧化活性的山桐子粕多酚冻干粉,采用DPPH自由基清除率、ABTS自由基清除率、总抗氧化能力、还原能力测定方法,评价山桐子粕多酚抗氧化活性.结果表明,各因素的影响顺序为乙醇体积分数>超声时间>料液比>超声温度.最佳提取工艺为乙醇体积分数65%,料液比1:60,超声温度63℃,超声时间110 min,山桐子粕多酚提取率为14.16%.山桐子粕多酚冻干粉对DPPH和ABTS自由基清除效果随着浓度的增加而持续上升,分别在1.6941 mg/mL和0.6128 mg/mL质量浓度时,其清除率大于同浓度的抗坏血酸对照.其还原能力与总抗氧化能力随浓度增加而增强,IC50分别为2.50704 mg/mL和2.67736 mg/mL,为山桐子粕多酚的高质化利用提供了一定的理论依据和新思路.
Optimization of Extraction Process of Polyphenols from Idesia polycarp Meal and in vitro Antioxidant Activity
Idesia polycarpa meal was taken as the research object,single factor experiments were conducted on ethanol concentration,material liquid ratio,ultrasonic temperature,and ultrasonic time.Response surface methodol-ogy was used to optimize the extraction process of polyphenols from idesia polycarpa meal,and the effects of various factors on the extraction rate of polyphenols from idesia polycarpa meal were evaluated.A certain antioxidant activity of polyphenols from idesia polycarpa meal freeze-dried powder was obtained through vacuum freeze-drying.The antioxidant activity was evaluated using DPPH free radical scavenging rate,ABTS free radical scavenging rate,total antioxidant capacity,and reducing capacity measurement methods.The results indicated that the order of influence of various factors was ethanol concentration>ultrasound time>material liquid ratio>ultrasound temperature.The opti-mal extraction process was ethanol concentration of 65%,solid-liquid ratio of 1:60,ultrasound temperature of 63℃,and ultrasound time of 110 minutes.Under these conditions,the extraction rate of polyphenols from idesia poly-carpa meal was 14.16% in three parallel experiments.The scavenging effect of polyphenols from idesia polycarpa meal freeze-dried powder on DPPH and ABTS free radicals continued to increase with the increase of concentration.At the concentrations of 1.6941 mg/mL and 0.6128 mg/mL,respectively,the scavenging rate was higher than that of the same concentration of vitamin C control.Its reducing ability and total antioxidant capacity increased with in-creasing concentration,with a concentration dependence.Within the concentration range of 0-5 mg/mL,respec-tively,the IC50 is 2.50704 mg/mL and 2.67736 mg/mL.The use of ultrasound assisted solvent extraction of poly-phenols from idesia polycarpa meal avoided resource waste and provided theoretical basis and new ideas for the high-quality utilization of polyphenols from idesia polycarpa meal,having certain reference value.
