基于数据挖掘与网络药理学的中医药治疗心肌缺血的用药规律及药理分析

目的 通过数据挖掘与生物信息学分析中医药治疗心肌缺血的用药规律及药理分析。方法 以"心肌缺血"作为主题词,收集相关文献并进行处方分析。将频率最高的药作为网络药理学分析对象,采用TC-MSP分析平台构建相应活性成分及靶点数据库,采用OMIM、TTD和Gencards数据库预测疾病相关的靶点基因。使用Venn取相应药组与疾病靶点的交集,将交集靶点导入STRING数据库构建蛋白相互作用网络,并将PPI网络导入Cytoscape 39。0软件筛选核心靶点,同时构建"药物-有效成分-靶点"相互作用网络。采用Metascape数据库进行GO富集分析和KEGG信号通路富集分析,并在微生信进行可视化处理。结果 共纳入符合要求的文献234篇,中医证型分布气虚血瘀证最多(56篇);高频药物分析,其中黄芪(159篇)、当归(138篇)、党参(127篇)的频数最高,用药靶向多集中于补虚、活血化瘀等,药性以温为主;药味以辛、甘最多;归经主要归属肝经、肺经和心经;在TCMSP数据库中筛选得到19个药物成分,根据度值排名槲皮素、木犀草素、芍药苷和异甘草素排名靠前;度值筛选出中药治疗心肌缺血排名前的核心靶点为磷脂酰肌醇3激酶(PIK3)、蛋白激酶(Akt-1)、白细胞介素6(IL-6)、血管内皮生长因子(VEGF),通过GO和KEGG的富集分析,分析出药物主要通过参与氧化应激、凋亡等生物过程以及凋亡、铁死亡等信号通路治疗心肌缺血。结论 中医药可能通过槲皮素、木犀草素等成分通过作用于PI3K、AKT1、IL-6、VEGF调节氧化应激反应、凋亡信号通路治疗心肌缺血。
Pharmacological analysis and medication rules of myocardial ischemia based on data mining and network pharma-cology
Objective To analyze the medication law and pharmacological analysis of traditional Chinese medi-cine in the treatment of myocardial ischemia through data mining and bioinformatics.Methods With"myo-cardial ischemia"as the theme word,the relevant literature was collected and the prescription was analyzed.The drugs with the highest frequency were taken as the objects of network pharmacology analysis.The tcmsp analysis platform was used to build the corresponding active ingredient and target database,and OMIM,TTD and gencards databases were used to predict the disease-related target genes.Venn was used to take the inter-section of the corresponding drug group and the disease target,and the intersection target was imported into the STRING database to construct the protein interaction network,and the PPI network was imported into Cytoscape 39.0 software to screen the core target,and the"drug-active ingredient-target"interaction network was constructed at the same time.The metascape database was used for go enrichment analysis and KEGG signaling pathway enrichment analysis,and the visualization processing was carried out in the wechat.Results Two hundred and thirty-four cases of eligible literature were included,and the distribution of TCM syn-drome types was Qi deficiency and blood stasis syndrome(56);According to the analysis of high-frequency drugs,Astragalus membranaceus(159),Angelica sinensis(138)and Codonopsis pilosula(127)have the highest frequency.The drug targets are mostly focused on tonifying deficiency,activating blood circulation and removing blood stasis,and the drug properties are mainly warm;Pungent and sweet herbs are the most popular;Meridians mainly belong to liver,lung and heart meridians;Nineteen pharmaceutical ingredients were screened in tcmsp database,and quercetin,luteolin,paeoniflorin and isoliquiritigenin ranked high ac-cording to the degree value;The core targets of traditional Chinese medicine in the treatment of myocardial ischemia were selected as PI3K,Aktl,IL6,VEGF by the enrichment analysis of go and KEGG pathways.The drug treatment of myocardial ischemia mainly involved in biological processes such as the response to ox-idative stress and apoptosis,as well as signaling pathways such as apoptosis signaling pathway and iron death signaling pathway.Conclusion Traditional Chinese medicine may act on PI3K,Aktl,IL6,VEGF through quercetin,luteolin and other components to regulate the response of oxidative stress and apoptotic signaling pathway in the treatment of myocardial ischemia.
