摘要
目的 探索细胞焦亡在肺鳞状细胞癌(LUSC)发生和发展中的作用,并基于生物信息学结果联合人群资料建立统计学预后模型,为临床诊断和治疗提供新策略.方法 采用癌症基因组图谱(TCGA)数据库,比较502例LUSC肿瘤组织和49例相邻非肿瘤肺组织的表达谱.运用Lasso Cox回归方法筛选与预后相关的差异表达基因开展生存、免疫细胞浸润等多维度分析.结果 基于11个细胞焦亡相关基因,成功构建出LUSC预后模型,相关差异表达基因主要参与了 IL-17信号通路、类风湿关节炎和百日咳等生物学过程.结论 本研究揭示了细胞焦亡在LUSC中的重要性,识别出一组关键基因,这些基因不仅可作为预后的生物标志物,而且为LUSC的个体化治疗提供了潜在靶点.
Abstract
Objective This study aims to explore the role of pyroptosis in the occurrence and development of Lung Squamous Cell Carcinoma(LUSC)and to establish a statistical prognostic model by integrating bioin-formatics results with population data,thereby offering new strategies for clinical diagnosis and treatment.Methods Utilizing the cancer genome atlas(TCGA)database,we compared the expression profiles of five hundred and two LUSC tumor tissues and 49 adjacent non-tumor lung tissues.The Lasso Cox regression method was employed to screen prognostically relevant differentially expressed genes for multidimensional a-nalysis,including survival and immune cell infiltration.Results A prognostic model for LUSC was success-fully constructed based on 11 pyroptosis-related genes.These differentially expressed genes were primarily involved in biological processes such as IL-17 signaling pathway,rheumatoid arthritis,and whooping cough.Conclusion This study underscores the significance of pyroptosis in LUSC,identifying a set of key genes.These genes not only serve as biomarkers for prognosis but also offer potential targets for personalized treat-ment of LUSC.
