摘要

为探讨不同解冻方法对纯种邓川牛公牛细管冻精品质的影响,本试验采用13℃、38℃、75℃三个不同的解冻温度对邓川牛细管精液解冻不同的时间,探索解冻后精子活力、有效生存时间以及顶体完整率变化,以期找到最合适的解冻方法.结果显示,在13℃解冻条件下解冻,精子的活率均低于0.3,38℃环境中解冻20s的精子活力较高,为0.43,75℃条件下解冻3s的精子活力最高,能达到0.45;不同温度解冻精子活率差异性显著(P<0.05).38℃和75℃解冻后精子存活时间随解冻温度的升高而降低,不同解冻温度解冻不同时间精子存活时间差异显著(P<0.05).75℃环境中解冻3s精子顶体完整率最高,在57%左右.在兼顾解冻效果与输精环境的基础上,可选用75℃3s解冻或38℃20s解冻.

Abstract

In order to explore the effects of different thawing methods on the quality of semen from Dengchuan cattle after thawing,three different thawing temperatures of 13 ℃,38 ℃ and 75 ℃ were used to thaw the semen of Dengchuan cattle at different times.The aim was to explore the sperm viability,effective survival time,and acrosome integrity rate after thawing,in order to find the most suitable thawing method.The results showed that when thawed at 13 ℃,the sperm viability was lower than 0.3,and the sperm viability was the highest at 75 ℃ for 3 seconds,reaching 0.45.The sperm viability was higher after thawing at 38 ℃ for 20 seconds,and there was a significant difference in sperm viability(P<0.05).After thawing at 38 ℃ and 75 ℃,the survival time of sperm decreases with the increase of thawing temperature,and there is a significant difference in the survival time of sperm thawed at different thawing temperatures and times(P<0.05).The highest acrosome integrity rate of thawed sperm for 3 seconds in 75 ℃ environment is around 57%.On the basis of considering the thawing effect and thawing environment,the semen quality of Dengchuan cattle fine tube frozen semen thawed at 75 ℃ for 3 seconds is higher.
