奶牛源多重耐药金黄色葡萄球菌耐药机制研究

本研究分析一株奶牛乳房炎源金黄色葡萄球菌(Staphylococcus aureus,S.aureus)Q1的耐药机制,为防控金黄色葡萄球菌性奶牛乳房炎提供数据基础.试验采用菌株分离鉴定、药敏试验及全基因组测序方法对其耐药机制进行预测.结果表明:Q1对大环内酯类药物(MA)、林可酰胺类药物(LA)、酰胺醇类药物(AMs)、四环素类药物(TCs)、磺胺类药物(SAs)均具有耐药性;对青霉素类药物(PCs)、氨基糖苷类药物(AGs)具有中介耐药性,且携带有ermB、ermC、msrA、mphC、lnuA、fexA、tetK、AAC(6')-Ie-APH(2')-Ia、CarB-6 9种耐药基因.说明,分离所得奶牛源S.aureus Q1具有多重耐药性,推测其耐药机制为外排泵、抗生素靶点改变及抗生素失活机制共同作用,可为S.aureus引起的奶牛乳房炎提供防治依据.
Study on Drug Resistance Mechanism of Multidrug Resistant Staphylococcus aureus Isolated from Dairy Cows
In order to analyze the drug resistance mechanism of a strain of Staphylococcus aureus(S.aureus)Q1 from cow mastitis,and provide data basis for the prevention and control of cow mastitis S.aureus.The drug resistance mechanism was predicted by strain isolation and identification,drug sensitivity test and whole genome sequencing.The results showed that Q1 was resistant to macrolides(MA),Lincosamides(LA),amidols(AMs),tetracyclines(TCs),and sulfonamides(SAs).It has intermediary resistance to penicillins(PCs)and aminoglycosides(AGS),and carries 9 resistance genes,including ermB,ermC,msrA,mphC,lnuA,fexA,tetK,AAC(6')-ie APH(2')-IA,carB-6.It is concluded that the isolated S.aureus Q1 from dairy cows has multiple drug resistance,and its drug resistance mechanism is presumed to be the combined action of efflux pump,antibiotic target change and antibiotic inactivation mechanism,which can provide a basis for the prevention and treatment of bovine mastitis caused by S.aureus.
