虎榛子组织培养及其菌根化技术研究

Studies on the Tissue Culture and Mycorrhizal Technology of Ostryopsis davidiana

为了获得大量的虎榛子组培幼苗,并对其进行瓶内菌根合成试验研究,以虎榛子带腋芽的茎段为外植体,进行外植体的灭菌,基础培养基的筛选,增殖培养和生根培养并对虎榛子组培幼苗进行瓶内菌根化研究.结果表明:经75%酒精处理30 s,5%NaClO处理7 min,虎榛子成活发芽率达到66%;WPM为最佳基础培养基;诱导虎榛子芽增殖的最佳培养基为WPM+6-BA 0.5 mg/L+NAA 0.03 mg/L,增殖率达到436.7%;最佳生根培养基为WPM+NAA 0.2 mg/L,生根率达到100.0%.对生根10天的虎榛子幼苗进行瓶内菌根合成试验,结果显示:接种土生空团菌[Cenococcum geophilum Fr.(Cg)]CgO5后虎榛子组培苗各项生长指标均显著提高,试验结果将为虎榛子幼苗快繁及其组培苗菌根化技术提供依据.
In order to get a number of plantlets and make mycorrhizal synthesis test of Ostryopsis davidiana, the Ostryopsis davidiana of which the stem segments with axillary buds were taken as the explants. The tissue culture from aspects of disposal, selection of basic medium, proliferation, rooting and mycorrhizal research of Ostryopsis davidiana were studied in the experiment. The results showed that: the best way of sterilization was soaked in 75% alcohol for 30 s, then treated in 5% NaClO for 7 min, then the germination rate could reached to 66%; the optimal basic medium was the WPM; the optimal proliferation medium for the buds was WPM+6-BA 0.5 mg/L + NAA 0.03 mg/L and the proliferation rate reached to 436.7%; for rooting, the optimal medium was WPM+NAA 0.2 mg/L. The rooting rate was up to 100.0%. Mycorrhizal synthesis test in the tissue culture bottle of Ostryopsis davidiana after rooting for 10 days showed that multiple growth indices of Ostryopsis davidiana plantlets were improved definitely after inoculated CgO5. The results provided the theoretical basis for the rapid propagation and mycorrhizal technology of Ostryopsis davidiana.
