Effects of Arbuscular mycorrhizal Fungi on Xylem Structure and Expression of Defense-related Genes in Salvia miltiorrhiza
To study the effects of GV,an Arbuscular mycorrhizal fungi,on the structure of xylem and synthesis related genes PAL and C4H,defense-related genes PR1,WRKY,the authors set up the control group and mycorrhizal group.After 90 days,the effects of mycorhiza on the Salvia miltiorrhiza growth and the structure of xylem were detected.The expressions of PAL,C4Hand PRIand WRKYwere recorded using real-time PCR at 30,60,75,90,105,and 130 days.The infection rate of S.miltiorrhiza root was 90%,which reached the peak basically.The fresh weight of the above-ground portion and the number of leave in the GV group were significantly higher than that of the control group,about 2.7 and 1.96 times.The integral optical density and area of xylem on root transection of GV group were significantly greater than that of the control group.The expression level of PAL and C4H in the roots was significantly increased in the GV group,while markedly decreased in the above-ground parts.The expression of PR1 in the roots and WRKY in both the roots and above-ground parts increased significantly before 75 days,and the expression of WRKYin roots saw another notable increase at 130 days.Arbuscular mycorrhizal fungi could promote growth,increase the area and density of xylem in the root of S.miltiorrhiza,and play a positive role in its pathogen resistance.
