Polygonatum sp.Transcriptome:SSR Markers Development and Genetic Diversity Analysis
To develop simple sequence repeat(SSR)markers for germplasm resource identification and genetic diversity analysis in Polygonatum sp.,transcriptome sequencing was conducted on 3 species of Polygonatum sp.,from which 19630 SSR loci containing 1-6 base repeats were identified out of 60836967 Unigenes.9 pairs of effective SSR primers were designed and screened,and 31 Polygonatum germplasm resources from 5 provinces were identified and subjected to genetic diversity analysis.The 9 pairs of SSR primers could not only distinguish P.filipes from other species of Polygonatum,but also effectively differentiate between different sources of Polygonatum(P.sibiricum,P.kingianum,and P.cyrtonema),with the genetic similarity coefficient of the 31 resources ranging from 0.0769 to 0.75.Additionally,a DNA fingerprint database for 31 germplasm resources of Polygonatum sp.was established,along with the development of the corresponding fingerprint maps.The results of this study provide useful molecular markers and a reference basis for the classification and identification,genetic diversity analysis,and variety selection of Polygonatum germplasm resources.
