Molecular identification and evolutionary analysis of cysticercus derived from Ochotona curzoniae in Qinghai Province
To identify the species of cysticercus derived from Ochotona curzoniae in Qinghai Province,the cysticercus samples were observed under a somatic microscopy,the mitochondrial COI gene sequences of cysticercus samples were amplified and sequenced,and the amplified products were compared.At the same time,the phylogeny and differentiation time of the sequenced COI genes were analyzed by Iqtree,BEAST v2.6.2 and other software.The results showed as follows:1)The morphological characteristics of the cysticercus were similar to those of Taeniidae cysticercus in the family Ceratopsidae.2)The sequencing results showed that the sequence was homologous to the gene sequence of Taenia caixuepengi in GenBank(MT882036),with a sequence similarity of 98.52%,indicating that the cysticercus belongs to Taenia caixuepngi.3)The phylogenetic tree showed that T.pisiformis was closely related to T.pisiformis,forming small branches between the two.4)The differentiation time analysis showed that T.caixuepengi and T.pisiformis differentiated at around 5.79Mya(95%confidence interval,3.15-6.87 Mya)with a bootstrap value of 100%.In conclusion,the cysticercus derived from O.curzoniae in Qinghai Province was identified as T.caixuepengi.Phylogenetic analysis and differentiation time analysis using the mitochondrial COI gene of this cysticercus showed that this cysticercus was more closely related to T.pisiformis.In this study,the identification and genetic evolution analysis were performed to provide theoretical basis for the classification,identification and molecular epidemiological investigation of T.caixuepengi in Qinghai province.
