蝴蝶兰'郑农红玉'组培快繁技术研究

为筛选一套简便高效的'郑农红玉'蝴蝶兰良种组培快繁技术体系,以满足市场的规模化生产需求,以蝴蝶兰新品种'郑农红玉'的花梗腋芽为外植体,依次进行腋芽诱导、丛生芽增殖、壮苗和生根培养,将基本培养基、植物生长调节剂和有机添加物等影响因素分别设为3个处理进行正交试验,以此探寻适宜不同生长阶段的培养基配方.结果表明:'郑农红玉'腋芽在"花宝1号"2g/L+"善存"多种维生素矿物质片1颗/L+肌醇125mg/L+6-BA 5.0mg/L+IBA 0.5mg/L+土豆泥1000g/L+蔗糖25g/L的培养基上诱导效果较好;切掉顶芽的芽苗在"花宝1号"2g/L+"善存"多种维生素矿物质片1颗/L+肌醇125mg/L+6-BA 2.5mg/L+IBA 0.5mg/L+土豆泥1000g/L+蔗糖20g/L的培养基上增殖倍数达到4倍;增殖苗在壮苗培养基"花宝1号"2g/L+"善存"多种维生素矿物质片1颗/L+肌醇125mg/L+IBA 0.5mg/L+NAA 0.7mg/L+土豆泥1000g/L+蔗糖25g/L中能达到整齐一致;无根苗在"花宝1号"2g/L+"善存"多种维生素矿物质片1颗/L+肌醇125mg/L+IBA 0.5mg/L+香蕉泥60g/L+土豆泥60g/L+AC 1g/L+蔗糖20g/L的培养基上生根效果最佳.研究结果可为规模化快繁蝴蝶兰'郑农红玉'提供技术支撑.
Research on Tissue Culture and Rapid Propagation Technology of Phalaenopsis'Zhengnong Hongyu'
In order to screen a set of simple and efficient tissue culture and rapid propagation technology system of P.'Zhengnong Hongyu',and meet the market demand for large-scale production,the axillary buds of the pedicel of P.'Zhengnong Hongyu'were used as explants.Pedicel axillary bud induction,proliferation of clustered buds,robust seedling and rooting cultures were carried out sequentially,and the influencing factors such as basic medium,plant growth regulators and organic additives were set as three treatments for orthogonal tests,so as to explore the suitable medium formulations for different growth stages.The results showed that pedicel axillary buds were better induced on the medium of"Hyponex NO.1"2 g/L+"Centrum"1 capsule/L+inositol 125 mg/L+6-BA 5.0 mg/L+IBA 0.5 mg/L+mashed potato 1000 g/L+sucrose 25 g/L;the shoots with the terminal buds cut off grew better on the medium of"Hyponex NO.1"2 g/L+"Centrum"1 capsule/L+inositol 125 mg/L+6-BA 2.5mg/L+IBA 0.5 mg/L+mashed potato 1000 g/L+sucrose 20 g/L,and the proliferation rate reached 4 times;the proliferated seedlings could reach uniformity in the seedling-strengthening medium of"Hyponex NO.1"2 g/L+"Centrum"1 capsule/L+inositol 125 mg/L+IBA 0.5 mg/L+NAA 0.7 mg/L+mashed potato 1000 g/L+sucrose 25 g/L;the rootless seedlings rooted best on the medium of"Hyponex NO.1"2 g/L+"Centrum"1 capsule/L+inositol 125 mg/L+IBA 0.5 mg/L+mashed banana 60 g/L+mashed potato 60 g/L+AC 1 g/L+sucrose 20 g/L.The aim was to lay a technical foundation for large-scale rapid propagation of P.'Zhengnonghongyu'.
