摘要

为了探究低聚半乳糖(GOS)植物乳杆菌发酵乳的潜在功能特性,通过在发酵乳中添加不同含量的GOS,对其差异代谢产物和代谢通路进行分析,对加入发酵乳培养的人体粪便肠道菌群进行物种α-多样性分析、群落的组成分析和物种β-多样性分析.结果表明,功能性差异代谢物主要为氨基酸、有机酸、核苷酸及碳水化合物,其中胞嘧啶核苷、异柠檬酸盐、乌头酸、肌醇、β-羟基丁酸含量增加,1-磷酸葡萄糖、尿酸含量降低;氨基糖和核苷酸糖代谢途径、半乳糖代谢途径、TCA循环、嘧啶代谢途径存在显著差异(P<0.05);添加GOS的植物乳杆菌发酵乳与空白对照组之间菌群组成相似性低,GOS植物乳杆菌发酵乳可以提升双歧杆菌(Bifidobacterium)、拟杆菌属(Bacteroides)等有益菌的相对丰度,抑制有害菌的繁殖,当GOS添加量为4%时,GOS植物乳杆菌发酵乳对肠道菌群的调节作用最有效.研究为进一步了解GOS植物乳杆菌发酵乳的功能性以及产品开发提供了理论参考与技术支持.

Abstract

To investigate the potential functional properties of Lactobacillus plantarum fermented milk added with galacto-oligosaccharide(GOS),different levels of GOS were added to the fermented milk,and their differential metabolites and metabolic pathways were analyzed,followed by species-diversity analysis,community composition analysis,and species-diversity analysis of human fecal intestinal flora added with the fermented milk.The results revealed that the functional differential metabolites were primarily amino acids,organic acids,nucleotides,and carbohydrates,with the contents of cytosine nucleoside,isocitrate,ursolic acid,inositol,and β-hydroxybutyric acid increasing and the contents of glucose 1-phosphate and uric acid decreasing;the amino and nucleotide sugar metabolic pathways,galactose metabolic pathways,TCA cycle and pyrimidine metabolic pathways were significantly different(P<0.05).The similarity of bacterial flora composition between the GOS fermented milk and blank control groups was low.GOS fermented milk could enhance the relative abundance of beneficial bacteria such as Bifidobacterium and Bacillus spp.and inhibit the propagation of harmful bacteria,and the regulation effect of GOS fermented milk on intestinal flora was greatest when the amount of GOS added was 4%.This study provides theoretical and technical assistance for further understanding the function of L.plantarum fermented milk with GOS and the product development.
