Study on the weight loss effect of Lactiplantibacillus plantarum J26 combination on high-fat diet-induced obese mice
Polysaccharides are a class of bioactive molecules from a wide range of sources that have the potential to regulate intestinal microbiota.In this study,the optimal fermentation conditions for exopolysaccharide of Lacticaseibacillusparacasei JY56 were determined using single-factor and response surface optimization experiment:the fermentation temperature was 30 ℃,the fermentation time was 26 h,and the fermentation pH was 6.0.A combina-tion of Lactiplantibacillusplantarum J26 and Lacticaseibacillus paracasei JY56 exopolysaccharide(J26&JY56 Exopolysaccharide Composition,JEC)was pre-pared.T o study the effect of JEC on obesity,a high-fat diet was used to establish an obese mouse model.After 10 weeks of dietary intervention,the results showed that JEC was effective in slowing down weight gain(P<0.05),alleviating liver fat accumulation due to chronic intake ofhigh-fat diet,and largely returning to normal levels.Specifically,the serum levels of TNF-α(301.77±9.87)ng/L,IL-6(225.39±10.09)ng/L,IL-1β(21.92±1.58)ng/L and IFN-γ(180.90±8.52)ng/L in the JEC group mice were significantly lower than those in the high fat diet induced obesity model mice group(HFD)(P<0.05).In addition,no significant lipid droplet vacuoles and necrotic cells were observed in the hepatocytes of the JEC group,and no inflammatory cell infiltration was observed.A combination ofLactiplantibacillusplantarum J26 and Lacticaseibacillus paracaseiJY56 exopolysaccharides had positive modulatory effects on delaying high-fat diet-induced obesity in mice.
