Isolation,identification and drug resistance analysis of Klebsiella pneumoniae from abnormal fresh milk
In order to find out whether there is Klebsiella pneumoniae infection and drug resistance gene in fresh milk produced by a dairy enterprise in southern Xinjiang,four abnormal fresh milk samples were isolated and cultured,16S rRNA gene sequence identification and mouse pathogenici-ty test were carried out by the routine pure culture method in the laboratory,and 16 antibiotics were selected to test the isolated strains with circu-lar disk diffusion method(K-B method)to detect the drug resistance gene.The results showed that 2 strains of Klebsiella pneumoniae were success-fully isolated;The isolated strain has multiple drug resistance The blaTEM,blaSHV and blaCTX-_M genes were detected in the strains resistant to β-lactam antibiotics;Only aac(6')-Ⅰb and oqxAB genes were detected in strains resistant to quinolones;Only sul1 and sul2 genes were detected in strains re-sistant to sulfa antibiotics;Among strains resistant to aminoglycoside antibiotics,only aac C2 and aph(3')-Ⅰa genes were detected;Among strains re-sistant to tetracycline antibiotics,only tet(A)gene was detected;It is highly pathogenic to mice,and the liver,spleen and lung of infected mice have different degrees of bleeding.The study showed that there was Klebsiella pneumoniae contamination in the raw and fresh milk produced by a dairy enterprise in southern Xinjiang.The test results provided a reference basis for enterprises to analyze the source of pollution,establish a scientific and accurate system for the prevention and control of microbial pollution,and lay a foundation for the public health and safety of raw and fresh milk in this area.
