Effects of Chito-oligosaccharides on the growth of probiotics in simulated intestinal medium
The objectice of this paper is to explore the effects of Chito-oligosaccharide on the growth of probiotics in stimulated intestinal fluid culture medium,so as to provide a theoretical basis for the development of probiotic and Chito-oligosaccharide synbiotics.Method:Eight probi-otics were inoculated into simulated intestinal fluid culture medium containing different concentrations of Chito-oligosaccharide for cultivation.The changes of cell density(△OD600),viable bacteria count,and the absorbance of culture at △OD260 nm were measured,as well as the integrity of cell membrane was characterized by scanning electron microscope.Results:all 8 probiotics grew normally in simulated intestinal fluid culture medium,and cannot grow with Chito-oligosaccharide as the sole carbon source.Chito-oligosaccharide inhibited the growth of 8 probiotics in simulation simulated intestinal fluid culture medium under natural cultivation process.Chito-oligosaccharide treatment led to surface collapse and wrinkles of probiotic cell membranes,disrupting the integrity of probiotic cell membranes.However,there was no unified inhibitory effect of Chi-to-oligosaccharide on the growth of 8 patented probiotics in alkaline simulated intestinal fluid culture medium.Conclusion:under alkaline simulat-ed intestinal fluid environment,Chito-oligosaccharide has a weak or no inhibitory effect on the proliferation of probiotics.Probiotics can be com-bined with Chito-oligosaccharide to develop synbiotics for regulating intestinal microecology.
