摘要

[目的]建立聚葡萄糖、甜菊糖苷中蛋白酶活力检测方法.[方法]通过单因素试验、正交试验优化蛋白酶活力的检测条件,考察回收率试验方法准确度.[结果]聚葡萄糖、甜菊糖苷中蛋白酶活力最佳检测条件:硼酸缓冲溶液溶解样品,待测样品取样量5 g,反应时间20 min,反应温度40℃,pH值10.5,滤液静置时间20 min.回收率试验结果表明,方法准确度64%~102%,相对标准偏差<1.3%.[结论]曲线相关性、准确度等验证,该方法适用聚葡萄糖、甜菊糖苷中蛋白酶含量检测.

Abstract

[Objective]The detection method of protease activity in polyglucose and stevioside glycosides was established.[Method]Single factor experiment and orthogonal experiment were used to optimize the assay conditions of protease activity,and the accuracy of the recovery method was investigated.[Result]The optimal conditions for the detection of protease activity in polyglucose and stevioside glycosides were to dissolve samples with boric acid buffer solution,sample size to be tested was 5 g,reaction time was 20 min,reaction temperature was 40 ℃,pH value was 10.5,and filtrate standing time was 20 min.The experimental results showed that the accuracy of the method was 64%~102% and the relative standard deviation was less than 1.3%.[Conclusion]This method was suitable for the detection of protease content in stevioside polyglucose after comprehensive verification of curve correlation and accuracy.
