基于TG-DSC联用法的腺苷钴胺热分解机制研究

目的 研究腺苷钴胺原料药的热分解规律,为其稳定性研究提供理论依据.方法 在采用X-PRD,IR和HPLC法对腺苷钴胺检测样品进行定性和定量分析的基础上,对相同粒度样品和不同粒度的样品进行TG-DSC检测,分析失重、峰形和峰值特征,得到腺苷钴胺热分解的规律,并讨论其影响因素. 结果 首次提出腺苷钴胺二次失水的热分解机制,即腺苷钴胺在64 ℃~83 ℃发生一次脱水,在206 ℃附近发生二次脱水,在276 ℃发生完全分解. 结论 该研究结果为腺苷钴胺原料药质量控制的开展提供了重要的理论研究依据.
Study on thermal decomposition mechanism of cobamamide based on TG-DSC analysis
Objective In order to provide the theoretical basis for the stability study of cobamamide, the thermal decomposition mechanism of it has been studied.Methods Based on the qualitative and quantitative analysis of cobamamide samples by X-PRD, IR and HPLC method, TG-DSC analysis of samples with same particle size and other samples with different particle size have been done.Weight loss, peak shape and peak characteristics have been compared, the decomposition of cobamamide thermal mechanism has been obtained and the influencing factors have been discussed.Results The thermal decomposition mechanism of second times dewatering of cobamamide was forward for the first time.The first dewatering occurs at 64 ℃-83 ℃, the second dewatering occurs at around 206 ℃, and complete decomposition occurs at around 276 ℃.Conclusion The results of this study provide important theoretical basis for the development of the quality control of cobamamide.
