The mutagenic effects of NTG on the protopasts from Aspergillus niger G27 was investigated in this study. The protoplasts were firstly produced by complex enzyme treatment of the mycelium,which were collected from slant cul-tures. After treatmented by 1 × 10-3 mmol/mL NTG for 0~60 min,the mortality mutation rate and the lipase activity of the stain was studied.The regeneration rate of protoplast was 7.5%.The mortality,mutation rate of protoplast increase along with the increase of NTG dose,and then became stable.Accroding to the increase of the NTG dose, the average lipase activity, standard deviation value and the coefficient of variation increased. After screening,two high yield and stable heredity strains was obtained,the P316 strain and the P388 stain,of which yield were(394.3 ± 12.9)U/mL with 22.9%higher than G27 and(385.1 ± 11.4)U/mL with 20.1%higher than G27 respectively.Protoplasts with NTG mutagenesis was demonstrat-ed to improve the efficiency of positive mutant,and have more stable heredity strain.
