地衣芽孢杆菌产芽孢培养基和培养条件的优化

本研究以地衣芽孢杆菌CICC 20514为对象,芽孢数和芽孢率为目标参数,采用单因素、正交试验方法 对培养基配方进行筛选,并在此配方基础上对培养条件进一步优化.结果 表明:最终产芽孢发酵培养基的最佳组合为20 g/L 果糖、20 g/L 豆粕、5 g/LK2HPO4、7 g/L NaCl、0.8 g/L CaCO3和 0.8 g/L MgSO4;产芽孢最佳培养条件为 37 ℃,220 r/min,接种量5%,装样量20%,初始pH 6.5~7.5,发酵培养时间48 h.优化后的地衣芽孢杆菌CICC 20514培养基芽孢数及芽孢率均具有良好的稳定性,发酵液芽孢数达5.8 × 109 CFU/mL,比优化前提高了 58倍.
Optimization on sporulation medium and culture conditions of Bacillus licheniformis
The medium formulation was screened by single-factor and orthogonal tests for Bacillus licheniformis CICC 20514 with the number and rate of spores as a screening condition,and the culture conditions were further optimized on the basis of this formulation.The results showed that the optimal combination of spore fermentation medium contains 20 g/L fructose,20 g/L soybean meal,5 g/L K2HPO4,7 g/L NaCl,0.8 g/L CaCO3 and 0.8 g/L MgSO4.The optimal culture conditions for spores production were 37 ℃,220 r/min,5% inoculum,20%sample loading,initial pH 6.5~7.5 and 48 h of fermentation.The optimized Bacillus licheniformis CICC 20514 medium had good stability in spore number and rate.The number of spores in the fermentation reached 5.8×109 CFU/mL,which was 58 times higher than before optimization.
