大理牛街热泉来源嗜热纤维素酶基因的克隆、异源表达及其酶学性质研究

纤维素作为自然界储量较大且廉价的可再生资源,其能被纤维素酶有效分解.由于工业生产常在高温下进行,所以嗜热纤维素酶成为研究重点.本研究从大理牛街热泉宏基因组测序数据中获得一个纤维素酶基因,命名为njcel6.将该基因在大肠杆菌中进行异源表达,并对其酶学性质进行研究.结果表明:该纤维素酶的最适pH为5.0,最适温度为50 ℃;50 ℃孵育120 min保持100%的相对活性,pH 3~10缓冲液中,孵育24 h仍能保持80%以上相对活性.该酶在K+、Mg+、Fe3+、Ni2+和Co2+等11种金属离子存在下均能保持85%以上相对活性.浓度为0.1%和1%的SDS抑制剂对酶活力抑制作用明显,抑制率达80%.综上所述,njcel6是一种具有良好热稳定性和酸碱稳定性的嗜热纤维素酶,该酶在食品加工和饲料生产等行业具有广泛的运用价值.
Cloning,heterologous expression and characterization of a thermophilic cellulase gene from Niujie hot spring in Dali
Cellulose,as a cheap and renewable resource in nature,can be decomposed by cellulase effectively.Thermophilic cellulase has become the focus of research,because industrial production is often carried out at high temperatures.In this study,a cellulase gene named njcel6,was obtained from the metagenomic sequencing data of Niujie hot spring in Dali.The gene was heterologous expressed in Escherichia coli and its enzymatic properties were studied.The results showed that the optimum pH and temperature of njcel6 were 5.0 and 50 ℃.After incubation at 50 ℃ for 120 min,it remained 100%relative activity,and it remained above 80%relative activity in pH 3~10 buffer after incubation for 24 h.In the presence of 11 metal ions such as K+,Mg2+,Fe3+,Ni2+and Co2+,it could maintain above 85%relative activity.The enzyme activity was inhibited significantly by 0.1%and 1%SDS and the inhibition rate reached 80%.In summary,njcel6 was a thermophilic cellulase with good thermal stability and acid-base stability,which had a wide range of application value in food processing and feed production.
