Cloning,heterologous expression and characterization of a thermophilic cellulase gene from Niujie hot spring in Dali
Cellulose,as a cheap and renewable resource in nature,can be decomposed by cellulase effectively.Thermophilic cellulase has become the focus of research,because industrial production is often carried out at high temperatures.In this study,a cellulase gene named njcel6,was obtained from the metagenomic sequencing data of Niujie hot spring in Dali.The gene was heterologous expressed in Escherichia coli and its enzymatic properties were studied.The results showed that the optimum pH and temperature of njcel6 were 5.0 and 50 ℃.After incubation at 50 ℃ for 120 min,it remained 100%relative activity,and it remained above 80%relative activity in pH 3~10 buffer after incubation for 24 h.In the presence of 11 metal ions such as K+,Mg2+,Fe3+,Ni2+and Co2+,it could maintain above 85%relative activity.The enzyme activity was inhibited significantly by 0.1%and 1%SDS and the inhibition rate reached 80%.In summary,njcel6 was a thermophilic cellulase with good thermal stability and acid-base stability,which had a wide range of application value in food processing and feed production.
