高效液相色谱-荧光法测定饲料中维生素B1和维生素B2

Determination of vitamin B1 and vitamin B2 in feed by HPLC with fluorescence

为建立以高效液相色谱-荧光法测定饲料中维生素B1和维生素B2的分析方法,实验通过高温高压法对饲料进行前处理,而后衍生,以乙酸钠-甲醇为流动相,分别在不同波长下对维生素B1和B2进行测定.结果显示:维生素B1和维生素B2在0.03~5.00 µg/mL线性范围内,相关系数R均大于0.9999;加标回收率为92.7%~98.1%;维生素B1和维生素B2的检出限分别为0.04、0.06 mg/kg,定量限分别为0.15、0.18 mg/kg.本方法对饲料中维生素B,和维生素B2的测定,具有便捷、准确的优势.
To establish an analytical method for the determination of vitamin B1 and vitamin B2 in feed by high performance liquid chromatography-fluorescence,the feed was pretreated by high temperature and high pressure method,and the extract was derivatized.Vitamin B1 and vitamin B2 were determined at different wavelengths with sodium acetate-methanol as mobile phase.The results showed that the liner range were 0.03~5.00 μg/mL.The correlation coefficients were greater than 0.9999.The recoveries were 92.7%~98.1%.The limit of detection respective were 0.04 mg/kg and 0.06 mg/kg of vitamin B,ang vitamin B2.The limit of quantitation respective were 0.15 mg/kg and 0.18 mg/kg of vitamin B1 ang vitamin B2.This method was convenient and accurate for the determination of vitamin B,and B2 in feed.
