Determination of sudan,rhodamine B and acid orange Ⅱ in poultry feed by high-performance liquid chromatography tandem mass spectrometry
This study was based on the high-performan ce liquid chromatography-tandem mass spectrometry(UPLC-MS)combined with QuEChERS rapid purification method,a method was developed for the quantitative analysis of sudan,rhodamine B and acid orange Ⅱ in poultry feed.The samples were extracted with acetonitrile and purified with C18+PSA.The purified liquid was separated by Hypersil GOLD C18 reversed-phase chromatography column,multi-reaction monitoring(MRM)was performed in an electrospray ionization source(ESI)with positive and negative ion double-scan mode,the quantity was determined by external standard curve method.The results showed that the correlation coefficient(R2)>0.999,the detection limits(LOD)of sudan and acid orange Ⅱ were 0.5 μg/kg and quantitative limits(LOQ)were 1.0 μg/kg,the LOD of rhodamine B were 0.05 μg/kg and the LOQ were 0.1 μg/kg.The recoveries ranged from 90%~110%,RSD<10%.This study had the advantages of fast and sensitive,it had realized the accurate quantitative analysis of 6 chemical dyes in poultry feed,and provided technical reference for the quality and safety of poultry feed.
