Optimization of extraction process of active peptide from Cordyceps militaris by response surface method
In order to optimize the extraction process of polypeptide from tussah silkworm pupa,the effects of pH,enzymatic hydrolysis time,reaction temperature and enzyme dosage were studied,and the extraction conditions were optimized by response surface method.The results showed that the effect order of four factors on the polypeptide yield of Cordyceps militaris cultivated with tussah pupa was pH>enzymatic hydrolysis time>reaction temperature>enzyme dosage.The optimum enzymolysis conditions were as follows:enzymolysis time 2.8 h,enzyme dosage 2%,pH 9.7,reaction temperature 56 ℃.The experiment was repeated three times under the condition of enzymolysis.The average yield of polypeptide was 86.1%,which was close to the predicted value of 87.52%,indicating that the model could well predict the assisted extraction of active peptide of Cordyceps militaris,and could provide a basis for the industrial development and application of Cordyceps militaris cultivated with tussah pupa.
