Effects of Scutellaria L.on PPARγ,APN and AMPK signaling pathways in the kidneys of obese hypertensive rats
This study was conducted to investigate the effect of Scutellaria L.root extract on fat deposition in rats and its mechanism.Twenty-four 8-week-old male SD rats were randomly divided into normal diet group and high-fat diet group, and each group was randomly divided into control group and Scutellaria L.root extract treatment group, namely control group(CTL),normal treatment group(TRT),high-fat group(F-CTL),high-fat treatment group(F-TRT),15 SD rats in each group. The TRT and f-trt groups were intragastrically administered with Scutellaria baicalensis root extract preparation every day,and CTL and f-ctl were intragastrically administered with the same amount of distilled water. After 10 weeks of treatment, the body weight, body fat rate, blood glucose, blood fat index,APN and PPAR of each group were compared and analyzed PPARγ,AMPK level and their mRNA expression level. The results showed that, for normal healthy rats, the treatment of Scutellaria L.root extract had no significant effect on body weight and lipid metabolism. However,in obesity, the former can activate PPARγ/AMPK signaling pathway by increasing adiponectin levels in adipose tissue and further promotes lipid catabolism, thereby reducing body weight, body fat rate and blood pressure in rats, and improving the increase of blood glucose caused by high-fat diet. Conclusion:Scutellaria L.root extract can activate APN/PPARγ/AMPK signaling pathway in adipose tissue, and plays a dual role in inhibiting obesity and lowering blood pressure.
