Effects of ambient temperature and dietary FagoPyrum dibotrys Hara extract on liver ultrastructure and mitochondrial antioxidant function of laying hens
The experiment aimed to investigate the effects of FagoPyrum dibotrys Hara extract(FDR)on the liver tissue ultrastructure and mitochondrial antioxidant function of Changshun green shell laying hens under different temperature conditions.192 laying hens were randomly into 12 groups with 4 replicates,each replicate 4 chickens.A 3×4 factorial design was adopted.Each process had 4 replicates,with 4 chickens per replicate.Arrange the experimental chickens into three independent temperature controlled chicken coops according to temperature gradients,and collect liver samples at the end of the experiment for ultrastructural and mitochondrial antioxidant index detection.The results showed that:(l)Under high temperature of 33 ℃,the liver tissue ultrastructure damage of laying hens was the most severe.Under high temperature treatment,the liver tissue ultrastructure damage was alleviated by adding different concentrations of FDR,while adding 400 mg/kg had the least damage.(2)Under 33 ℃ environmental conditions,glutathione peroxidase(GSH-Px)activity and superoxide dismutase(SOD)activity decreased by 13%and 6.1%,respectively,while malondialdehyde(MDA)content and protein carbonyl(PC)content increased by 17.3%and 13.9%,respectively.The effect of FDR addition on temperature showed an opposite trend.The addition of environmental temperature and FDR had a significant interaction effect on GSH-Px activity,SOD activity,MDA content and PC content in laying hens(P<0.05);(3)Adding FDR to the diet upregulated the mRNA expression of Nrf2 signaling pathway and Trx2 system related genes in the liver of laying hens,but not significantly(P>0.05).The addition of environmental temperature and FDR had a significant interaction effect on the relative mRNA expression of various genes in the liver of laying hens(P<0.05).In summary,long-term exposure to high temperature reduces the antioxidant capacity of the liver of laying hens.Adding 400 mg/kg or higher content of buckwheat extract to the diet could alleviate the ultrastructural damage of the liver tissue of laying hens under high temperature,and improve the antioxidant capacity of the liver mitochondria of laying hens.
