摘要

本研究从实验室构建的微生物菌库中筛选获得1株β-葡萄糖苷酶产生菌株HHL,结合ITS序列分析方法鉴定其为米曲霉(Aspergillus oryzae).以β-葡萄糖苷酶酶活为考察指标,通过单因素试验及正交试验对其产酶条件进行优化.结果显示:菌株HHL最优产酶条件为以改良察氏培养基为培养基,pH 6.0,接种量8%,培养温度35 ℃,培养时间144 h.此条件下,产β-葡萄糖苷酶酶活为48.74 U/mL.

Abstract

In this study,a strain named HHL with β-glucosidase production was screened from mycelium bank of the laboratory,and was identified as Aspergillus oryzae by ITS sequence analysis.The enzymatic activity of β-glucosidase as response value,the fermentation conditions were optimized by single factor and orthogonal tests.The results showed that the optimum conditions of β-glucosidase activity of HHL were as follows:the modified Czapek's medium was used as the fermentation medium,fermentation time was 144 h,fermentation temperature was 35 ℃,inoculums size was 8%,pH was 6.0.Under the optimal conditions,the β-glucosidase activity was 48.74 U/mL.
