植物多糖对长江鲟幼鱼不同组织抗氧化指标和免疫指标的影响及机理初探

为探究黄芪多糖(APS)和茯苓多糖(PCP)对长江鲟幼鱼不同组织器官抗氧化能力和免疫能力的影响,试验将90尾长江鲟幼鱼随机分为对照组、黄芪多糖组和茯苓多糖组,每组3个重复,每个重复10尾试验鱼,在基础日粮中按照体质量添加黄芪多糖和茯苓多糖,采用灌服方式投喂,试验周期为20 d.试验结束时每个试验组随机选取15尾鱼采集血清、肝脏、肾脏、肌肉、肠道、体表黏液等组织器官,采用双抗体夹心法检测抗氧化指标和免疫指标的变化情况,并分析植物多糖对长江鲟幼鱼不同组织器官抗氧化和免疫指标的影响程度.结果表明:(1)与对照组相比,黄芪多糖可显著增强长江鲟幼鱼血清、肝脏、肾脏、肠道、肌肉等组织中超氧化物歧化酶(SOD)的活性(P<0.05),提高率分别为43.36%、60.27%、43.75%、64.65%和56.87%;可显著增强血清、肝脏、肠道、体表黏液等组织中过氧化氢酶(CAT)的活性(P<0.05),提高率分别为27.97%、38.83%、126.43%和47.93%;可显著增强血清、肾脏、肌肉等组织中过氧化物酶(POD)的活性(P<0.05),提高率分别为12.39%、22.80%和14.14%;可显著增强血清、肝脏、肾脏、肠道、肌肉、体表黏液等组织中谷胱甘肽过氧化物酶(GSH-Px)的活性(P<0.05),提高率分别为24.34%、33.96%、34.65%、25.00%、15.73%和38.96%;可显著增强血清、肝脏、肾脏、肌肉、体表黏液等组织中总抗氧化能力(T-AOC)(P<0.05),提高率分别为16.88%、20.36%、10.59%、12.07%和13.56%;可显著降低血清、肝脏、肾脏、肠道、肌肉、体表黏液等组织中丙二醛(MDA)的含量(P<0.05),降低率分别为29.14%、25.18%、18.15%、28.53%、22.20%和24.40%.茯苓多糖可显著增强长江鲟幼鱼肠道、肌肉等组织中SOD的活性(P<0.05),提高率分别为71.95%和43.00%;可显著增强肾脏、体表黏液等组织中CAT的活性(P<0.05),提高率分别为23.25%和51.29%;可显著增强血清中POD的活性(P<0.05),提高率为16.87%;可显著增强血清、肾脏、体表黏液中GSH-Px的活性(P<0.05),提高率分别为21.78%、19.69%和36.81%;可显著增强血清、肾脏、肠道、体表黏液等组织中T-AOC(P<0.05),提高率分别为11.69%、13.24%、8.23%和10.17%;可显著降低血清、肠道等组织中MDA的含量(P<0.05),降低率分别为29.86%和24.03%.(2)与对照组相比,黄芪多糖可显著增强长江鲟幼鱼血清、肝脏等组织中IgG的含量(P<0.05),提高率为34.23%和39.39%;可显著增强血清、肾脏等组织中IgM的含量(P<0.05),提高率为23.16%和37.13%;可显著增强血清中C3、C4的含量(P<0.05),提高率分别为38.24%和27.88%;可显著增强肾脏组织中IFN-γ的含量(P<0.05),提高率为41.04%;可显著增强血清中碱性磷酸酶(AKP)的活性(P<0.05),提高率为25.99%;可显著增强血清、肝脏、肾脏、肠道、体表黏液等组织中溶菌酶(LSZ)的活性(P<0.05),提高率分别为59.75%、50.00%、75.00%、37.50%和66.50%;可显著增强血清、肝脏、肾脏、肠道、体表黏液等组织中抗菌肽(TP)的含量(P<0.05),提高率分别为39.37%、34.65%、21.24%、55.42%和36.38%.茯苓多糖可显著增强长江鲟幼鱼血清组织中IgG的含量(P<0.05),提高率为26.59%;可显著增强血清组织中C3的含量(P<0.05),提高率为44.50%;可显著增强血清、肝脏、肾脏、肠道等组织中LSZ的活性(P<0.05),提高率分别为36.55%、33.33%、25.00%和25.00%;可显著增强肠道、体表黏液等组织中TP的含量(P<0.05),提高率分别为36.56%和27.23%.由此得出,黄芪多糖和茯苓多糖均对长江鲟幼鱼不同组织器官的抗氧化和免疫机能具有改善作用,黄芪多糖作用更加显著.
Preliminary exploration on the effects and mechanisms of plant polysaccharides on antioxidant and immune indexes of different tissues of juvenile Acipenser dabryanus
To investigate the effects of Astragalus polysaccharides and Poria cocos polysaccharides on the antioxidant and immune abilities of different tissues and organs in juvenile Acipenser dabryanus.In the experiment,90 young Acipenser dabryanus fish were randomly divided into a control group,APS group,and PCP group.Each group had 3 replicates,with 10 experimental fish per replicate.Astragalus polysaccharides and Poria cocos polysaccharides were added to the basic diet according to their body weight,and were fed by gavage.The experimental period was 20 days.At the end of the experiment,15 experimental fish were randomly selected from each experimental group to collect tissues or organs such as blood,liver,kidneys,muscles,intestines,and body surface mucus.The double antibody sandwich method was used to detect antioxidant and immune indicators,and the impact of plant polysaccharides on the antioxidant and immune indicators of different tissues and organs of juvenile Acipenser dabryanus was analyzed.The results showed that:(1)Compared with the control group,Astragalus polysaccharides significantly enhanced the activity of SOD in tissues such as serum,liver,kidney,intestine,and muscle of juvenile Acipenser dabryanus(P<0.05),with an increase rate of 43.36%,60.27%,43.75%,64.65%,and 56.87%,respectively;It could significantly enhance the activity of CAT in tissues such as serum,liver,intestine,and body surface mucus(P<0.05),with an increase rate of 27.97%,38.83%,126.43%,and 47.93%,respectively;It could significantly enhance the activity of POD in tissues such as serum,kidneys,and muscles(P<0.05),with an increase rate of 12.39%,22.80%,and 14.14%,respectively;It could significantly enhance the activity of GSH-Px in tissues such as serum,liver,kidneys,intestines,muscles,and body surface mucus(P<0.05),with an increase rate of 24.34%,33.96%,34.65%,25.00%,15.73%,and 38.96%,respectively;It could significantly enhance T-AOC in tissues such as serum,liver,kidneys,muscles,and body surface mucus(P<0.05),with an increase rate of 16.88%,20.36%,10.59%,12.07%,and 13.56%,respectively;It can significantly reduce the content of MDA in tissues such as serum,liver,kidneys,intestines,muscles,and body surface mucus(P<0.05),with a reduction rate of 29.14%,25.18%,18.15%,28.53%,22.20%,and 24.40%,respectively.Poriacocos polysaccharides could significantly enhance the activity of SOD in the intestines,muscles and other tissues of juvenile Acipenser dabryanus(P<0.05),with an increase rate of 71.95%and 43.00%,respectively;It could significantly enhance the activity of CAT in tissues such as kidneys and body surface mucus(P<0.05),with an increase rate of 23.25%and 51.29%,respectively;It could significantly enhance the activity of POD in serum(P<0.05),with an increase rate of 16.87%;It could significantly enhance the activity of GSH-Px in serum,kidneys,and body surface mucus(P<0.05),with an increase rate of 21.78%,19.69%,and 36.81%,respectively;It could significantly enhance T-AOC in tissues such as serum,kidneys,intestines,and body surface mucus(P<0.05),with an increase rate of 11.69%,13.24%,8.23%,and 10.17%,respectively;It could significantly reduce the content of MDA in serum,intestinal and other tissues(P<0.05),with a reduction rate of 29.86%and 24.03%,respectively.(2)Compared with the control group,Astragalus polysaccharides could significantly enhance the IgG content in the serum,liver and other tissues of juvenile Acipenser dabryanus(P<0.05),with an increase rate of 34.23%and 39.39%;It could significantly enhance the content of IgM in serum,kidney and other tissues(P<0.05),with an increase rate of 23.16%and 37.13%;It could significantly enhance the content of C3 and C4 in serum(P<0.05),with an increase rate of 38.24%and 27.88%,respectively;It could significantly significantly enhance IFN-γ in kidney tissue(P<0.05),with an increase rate of 41.04%;It could significantly enhance the activity of AKP in serum(P<0.05),with an increase rate of 25.99%;It could significantly enhance the activity of LSZ in tissues such as serum,liver,kidneys,intestines,and body surface mucus(P<0.05),with an increase rate of 59.75%,50.00%,75.00%,37.50%,and 66.50%,respectively;It could significantly enhance the content of TP in tissues such as serum,liver,kidneys,intestines,and body surface mucus(P<0.05),with an increase rate of 39.37%,34.65%,21.24%,55.42%,and 36.38%,respectively.Poriacocos polysaccharides could significantly enhance the IgG content in the serum tissue of juvenile Yangtze sturgeon(P<0.05),with an increase rate of 26.59%;It could significantly enhance the content of C3 in serum tissue(P<0.05),with an increase rate of 44.50%;It can significantly enhance the activity of LSZ in tissues such as serum,liver,kidneys,and intestines(P<0.05),with an increase rate of 36.55%,33.33%,25.00%,and 25.00%,respectively;It can significantly enhance the content of TP in tissues such as intestines and body surface mucus(P<0.05),with an increase rate of 36.56%and 27.23%,respectively.From this,it could be concluded that both Astragalus polysaccharides and Poriacocos polysaccharides had an improvement effect on the antioxidant and immune functions of different tissues and organs in juvenile Acipenser dabryanus,with Astragalus polysaccharides having a more significant effect.
