Study on detection methods of four natural yellow pigments in chicken belly fat
To determine the content of four different natural pigments in chicken belly fat by HPLC,and to analyze the pigment components of five different sources of chicken belly fat.Samples were extracted with 0.5%BHT of methylene chloride/acetonitrile(3∶2,V/V)mixed solvent,10%anhydrous sodium sulfate solution and n-hexane separation,organic phase merged and concentrated,n-hexane dissolution and constant volume,part of the solution was added to 2 mL of 40%potassium hydroxide methanol solution vortex soap.After 1ml acetic acid acidification,10%anhydrous sodium sulfate solution and n-hexane were separated.The organic phase was merged and concentrated,and n-hexane was dissolved.The solution before and after saponification was dehydrated by anhydrous sodium sulfate and 0.22 μm organic filter membrane was used for machine detection.The liquid phase was equal elution of n-hexane/ethyl acetate/isopropyl alcohol(730∶270∶15,V/V/V)at the flow rate of 1.4 mL/min and wavelength of 445 nm.The results showed that the linear range of this method was 0.31~1.57 µg/mL,R2=0.9999,RSD were 2.68%~4.81%,and the detection limit was 0.05 mg/kg.When the dosage of lutein was 0.8~3.3 mg/kg,the recoveries were 96.74%~109.10%.The method could simultaneously detect the content of four different yellow pigments in chicken belly fat,with high sensitivity and good reproducibility,and provide technical support for the study of the coloring rule of yellow pigment in chicken belly fat.
