Study on the optimization of extraction conditions for biogenic amine in fish meal with sulfosalicylic scid
The aim of this experiment was to optimize the extraction conditions of three biogenic amines in fish meal with sulfosalicylic acid.Single factor experiment and orthogonal analysis were used to investigate the influence of four factors on the extraction effect of biogenic amine,namely,the type of extraction solution,weighing quality,concentration of extraction solution,and ultrasonic time.For the optimized conditions,the spiked recovery rate experiment and quality control sample experiment were carried out to verify the accuracy of the results.The results showed that the optimum extraction conditions of biogenic amine were selected by orthogonal experiment as follows:weighing 0.5~1.0 g,concentration of sulfosalicylic acid 1.0%,ultrasonic time 30 min.The recovery rate of the spiked sample was 101%~105%.The measured value of the quality control sample was 458 mg/kg,and the specified value was(452.8±64.9)mg/kg.The results were reliable.The biogenic amines in fish meal extracted by sulfosalicylic acid,ninhydrin were used as derivative reagents and detected by amino acid analyzer,which achieved good detection results and provided a simple and fast detection method.
