黄芪不同方法提取液对羔羊生长性能、血清细胞因子及抗氧化指标的影响

为研究不同提取方法取得的黄芪提取液对蒙古羊羔羊生长性能、血清细胞因子及抗氧化指标的影响,试验随机选取体重(22.64±1.21)kg、体尺和月龄相近的健康蒙古羊羔羊60只,分为对照组、黄芪水煎液组、黄芪超声液组和乳酸菌发酵黄芪水提液组,每组3个重复,每个重复5只.预试验10 d,正式试验60 d,于正式试验第60天静脉采血,分离血清并测定相关指标.结果显示:(1)与对照组相比较,Ⅰ组和Ⅱ组终末体重差异不显著(P>0.05),Ⅲ组终末体重比对照组、Ⅰ组、Ⅱ组分别增加4.5、3.71、2.46 kg(P<0.05);Ⅰ组、Ⅱ组和Ⅲ组平均日增重比对照组分别增加9.4、11.43、27.56 g/只(P<0.05);Ⅰ组、Ⅱ组和Ⅲ组料重比较对照组降低0.57、0.95和1.68(P<0.05).(2)与对照组相比,Ⅰ组白介素-6(IL-6)、白介素-10(IL-10)、白介素-12(IL-12)、肿瘤坏死因子α(TNF-α)和γ干扰素(IFN-γ)的含量分别降低了1.1、1.53、0.87、1.49 pg/mL 和 1.07 pg/mL(P<0.05),Ⅲ 组与 Ⅱ 组相比较,IL-10、TNF-α 和IFN-γ 的含量分别降低 0.78、1.14 pg/mL 和 0.45 pg/mL(P<0.05);Ⅱ 组 IL-6、IL-10 和 IL-12 含量比 Ⅰ 组降低0.5、0.67 pg/mL和0.72 pg/mL(P<0.05).(3)与对照组相比较,Ⅲ组谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)和超氧化物歧化酶(SOD)含量分别升高了2.13、2.81 U/mL 和 1.79 U/mL(P<0.05),Ⅲ组 GSH-Px、T-AOC和SOD含量比Ⅱ组分别升高了1.17、1.54 U/mL和0.9 U/mL(P<0.05).Ⅲ组过氧化氢酶(CAT)含量比照组、Ⅰ组和Ⅱ组分别升高了2.87、2.84 U/mL和2.37 U/mL(P<0.05).Ⅲ组血清中丙二醛(MDA)含量比对照组、Ⅰ组和Ⅱ组分别降低了2.86、2.54 nmol/mL和1.93 nmol/mL(P<0.05).综上所述,黄芪不同方法提取液对羔羊生长性能、血清细胞因子及抗氧化指标的影响不同,发酵黄芪水提液组效果更明显.
Effects of different extracts of Astragalus membranaceus on growth performance,serum cytokines and antioxidant indices of lambs
To study the effects of Astragalus extracted by different extraction methods on growth performance,serum cytokines and antioxidant indexes of Mongolian lambs,sixty healthy Mongolian lambs with similar body weight(22.64±1.21)kg,body size and month age were randomly selected and divided into control group,Astragalus decoction group,Astragalus ultrasonic solution group and fermented Astragalus extract group with 3 replicates per group and 5 lambs per replicate.The pre-test lasted for 10 days and the formal test lasted for 60 days.On the 60th day of the formal test,blood samples were collected intravenously,serum was separated and relevant indexes were determined.The results showed as follows:(1)Compared with the control group,the final body weight of groups Ⅰ and Ⅱ had no significant difference(P>0.05),and the final body weight of group Ⅲ was increased by 4.5,3.71 kg and 2.46 kg compared with that of control group,groups Ⅰand Ⅱ(P<0.05).The average daily gain in groups Ⅰ,Ⅱ and Ⅲ was 9.4,11.43 kg and 27.56 kg higher than that in control group,respectively(P<0.05).The ratio of feed to gain in groups Ⅰ,Ⅱ and Ⅲ was decreased by 0.57,0.95 and 1.68 compared with the control group(P<0.05).(2)Compared with the control group,the levels of interleukin-6(IL-6),interleukin-10(IL-10),interleukin-12(IL-12),tumor necrosis factor(TNF-a)and interferon-γ(IFN-γ)in group Ⅰ were reduced by 1.1,1.53,0.87,1.49 pg/mL and 1.07 pg/mL(P<0.05).Compared with group Ⅱ,the contents of IL-10,TNF-γ and IFN-γ in group Ⅲ were reduced by 0.78,1.14 pg/mL and 0.45 pg/mL,respectively(P<0.05).The contents of IL-6,IL-10 and IL-12 in group Ⅱ were decreased by 0.5,0.67 pg/mL and 0.72 pg/mL compared with group Ⅰ(P<0.05).(3)Compared with the control group,the contents of glutathione peroxidase(GSH-Px),total antioxidant capacity(T-AOC)and superoxide dismutase(SOD)in group Ⅲ were increased by 2.13,2.81 U/mL and 1.79 U/mL,respectively(P<0.05).Compared with group Ⅱ,the contents of GSH-Px,T-AOC and SOD in group Ⅲ were increased by 1.17,1.54 U/mL and 0.9 U/mL,respectively(P<0.05).The content of catalase(CAT)in groupⅢ was increased by 2.87,2.84 U/mL and 2.37 U/mL,respectively,compared with that in groups Ⅰ and Ⅱ(P<0.05).Serum malondialdehyde(MDA)content in group Ⅲ was decreased by 2.86,2.54 nmol/mL and 1.93 nmol/mL compared with control group,group Ⅰ and group Ⅱ,respectively(P<0.05).In conclusion,different extraction methods of Astragalus had different effects on growth performance,serum cytokines and antioxidant indexes of lambs,and the effects of fermented Astragalus water extract group were more obvious.
