枯草芽孢杆菌发酵蟹壳粉产蛋白酶的研究

本试验由蟹壳粉和蔗糖组成蟹壳粉培养基,通过测定发酵液中的蛋白酶活来确定最佳的培养基组成成分,利用单因素试验和响应面优化的方法优化蟹壳粉培养基,使枯草芽孢杆菌发酵蟹壳粉来产蛋白酶,提高废弃蟹壳的回收率,减少对环境的污染.试验结果表明:当蟹壳粉的浓度为5.12%,枯草芽孢杆菌的接种量为3.02%,蔗糖浓度为11.00%,初始pH为7.07时,枯草芽孢杆菌发酵蟹壳粉产酶的酶活最大值为17.4220 U/mL,较单因素试验时的最高蛋白酶酶活力14.2290 U/mL有一定的提升.
Study on protease production from crab shell meal fermented by Bacillus subtilis
This experiment consists of crab shell powder and sucrose as a crab shell powder medium.The optimal medium composition was determined by measuring the protease activity in the fermentation broth,and the crab shell powder medium was optimized using single factor text and response surface optimization methods.Make Bacillus subtilis ferment crab shell powder to produce protease,improve the recovery rate of waste crab shell,reduce environmental pollution.The experimental results showed that when the concentration of crab shell powder was 5.12%,the inoculation amount of Bacillus subtilis was 3.02%,the concentration of sucrose was 11%,and the initial pH was 7.07,the enzyme activity of Bacillus subtilis fermented crab shell powder was the largest.The value was 17.4220 U/mL,which was a certain increase compared with the highest protease activity of 14.2290 U/mL in the single factor text.
