Research of the mechanism on inflammatory injury of chicken heart induced by different concentrations of cadmium
To investigate the effects of different concentrations of cadmium(Cd)on heart damage and inflammation in chickens with chronic poisoning,a subchronic Cd poisoning model was established in Hailan-white chickens.Cd was added to feed to induce heart inflammatory damage in chickens.The study was divided into three groups:control group,35 mg/kg group and 70 mg/kg group,which were fed continuously for 90 days.After feeding,the heart was taken and cardiac histopathology,cardiac function indicators,serum inflammation indicators,and NOS enzyme activity were tested.The results showed that different concentrations of Cd led to changes in the morphology and pathological tissue structure of the chicken heart.Significantly increased of inflammatory infiltration of the heart and abnormal cardiac function indicators were obviously.Moreover,Cd caused an increase in cardiac Nitric oxide synthases(NOS)enzyme activity,total NOS(T-NOS)and inducible NOS(iNOS)activities(P<0.01,P<0.001),while constitutive NOS(cNOS)activity was not affected by Cd.In addition,Cd triggered significantly increased inflammatory cytokines of interlekin(IL)-6 and tumor necrosis factor alpha(TNF-α),IL-8,nuclear factor-κB(NF-κB)in mRNA levels in heart tissue(P<0.05,P<0.01,P<0.001),with a similar increase of content of serum IL-6,TNF-α and IL-1β(P<0.05,P<0.01).In conclusion that Cd could activate the expression and secretion of inflammatory factors and led to cardiac dysfunction and inflammatory damage in chickens.
