大孔树脂分离纯化黑穗醋栗叶片总黄酮的研究

黑穗醋栗叶片中含有丰富的黄酮类物质,此类物质具有抗肿瘤、抗氧化、抗菌消炎等多种功效,是一类非常重要的天然产物.本文采用6种大孔树脂(AB-8、NKA-9、D101-1、X-5、D4006、D4020)对从黑穗醋栗叶片中得到的黄酮粗提液进行分离纯化,通过对大孔树脂的静态吸附及解吸的研究,初步筛选出吸附和解吸能力较好的3种大孔树脂(NKA-9、D4020、D101-1).继续进行大孔树脂分离纯化黑穗醋栗叶片总黄酮吸附及解吸动力学和吸附等温线的试验,最终确定大孔树脂分离纯化黑穗醋栗叶片中总黄酮的最佳条件:在25℃下,使用NKA-9大孔树脂,纯化浓度为0.9311mg/mL的黄酮粗提液,体积60 mL,流速1.0 mL/min;洗脱剂(无水乙醇)用量100 mL,流速1.0 mL/min.用Sephadex LH-20凝胶分离得到的黄酮化合物组份Ⅰ,通过高效液相色谱测定并计算得到标准曲线,测得纯度为86.37%.
Study on the separation and purification of total flavonoids from black currant leaves by macroporous resins
Black currant leaves contain a variety of flavonoids,which has many functions,such as anti-tumor,anti-oxidation,anti-bacterial and anti-inflammatory.In this paper,six kinds of macroporous resins (AB-8,NKA-9,D101-1,X-5,D4006,D4020) were used to separate and purify flavonoids crude extract from the leaves of black currant.Based on the study of the static adsorption and desorption of macroporous resin,three kinds of macroporous resins (NKA-9,D4020,D101-1) were selected.Then the kinetics and adsorption isotherm for separation and purification were studied.The best conditions were determined as:temperature 25℃,NKA-9 macroporous resin,flavonoids concentration 0.9311 mg/mL,sample volume 60 mL,flow rate 1 mL/min;the eluent (absolute ethyl alcohol) dosage 100 mL,flow rate 1 mL/min.The flavonoids compound I was isolated by Sephadex LH-20 gel.The standard curve was obtained by high performance liquid chromatography and the purity was 86.37%.
