Purification of proanthocyanidins from Lycium ruthenicum Murr.by AB-8 macroporous resin and its stability
AB-8 macroporous resin was used to purify proanthocyanidins from Lycium ruthenicum Murr..The optimized purification parameters after static adsorption and desorption tests were:sample concentration 1mg/mL,eluent concentration70%,sample flow rate 2 mL/min,elution amount 5 BV,elution flow rate 2 mL/min,eluent amount 6 BV.Under these conditions,the purity of proanthocyanidins was (62.36 ± 0.76) %,and the recovery rate was (67.43 ± 1.48) %.Proanthocyanidins were sensitive to light and temperature,the stability was poor under strong light and high temperature.Oxidantive and reducing agent had significant effect on the stability;ions such as Fe3+,A13+,Zn2+,and Cu2+ can decrease the stability,while Na+,K+,Mg2+and Ca2+ had no significant effect;pH value had a great influence and sucrose,VC,citric acid and potassium sorbate had no significant effect on the stability.
