以雪菊为原料,采用水、30%、50%、70%、90% 的乙醇分别提取后,合并各提取液制得雪菊提取物,并对其进行体外抑菌活性试验,在此基础上,将其灌胃于慢性不可预知性温和应激(Chronic unpredictable mild stress,CUMS)抑郁模型小鼠,探究雪菊提取物对CUMS抑郁模型小鼠4种肠道菌的影响.结果显示,雪菊提取物总黄酮含量为30.00%、马里苷含量为0.83%,该提取物对大肠杆菌、沙门氏菌及金黄色葡萄球菌均有明显的抑制作用,其最低抑菌浓度分别为0.625、0.625、0.3125 mg/mL;CUMS抑郁模型小鼠给予雪菊提取物以后,不仅逆转了CUMS所致的小鼠抑郁行为,同时能够抑制肠球菌及大肠杆菌的生长繁殖、促进乳酸杆菌及双歧杆菌的生长繁殖,提高肠道定植力(B/E).综上所述,雪菊提取物不仅具有良好的体外抑菌能力,还能改善CUMS抑郁模型小鼠抑郁行为以及肠道菌的生长情况.
In vitro antibacterial activity of Coreopsis tinctoria Nutt. Extract and its effects on four kinds of intestinal bacteria in CUMS depression model mice
Coreopsis tinctoria Nutt. extracts were prepared using water and ethanol at concentrations of 30%,50%,70%,and 90%,followed by the combination of the extracts to evaluate the in vitro antibacterial activity. Subsequently,the extract was administered via gavage to mice subjected to chronic unpredictable mild stress (CUMS) to investigate its effects on four types of intestinal bacteria. The results indicated that the total flavonoid content of the combined extract was 30.00%,with a malicin content of 0.83%. In vitro analysis demonstrated the extracts exhibited significant antibacterial activity against Escherichia coli,Salmonella,and Staphylococcus aureus,with minimum inhibitory concentrations of 0.625,0.625,and 0.3125 mg/mL,respectively. Administration of the extract to CUMS depression model mice not only reversed the depressive behaviors induced by stress but also inhibited the growth of Enterococcus and E. coli,while promoting the proliferation of Lactobacillus and Bifidobacterium,thereby enhancing gut colonization (B/E ratio). In conclusion,Coreopsis tinctoria Nutt. extract demonstrated effective in vitro antibacterial properties and positively influenced depressive behaviors and gut microbiota in CUMS depression model mice.
