HPLC Determination of ATP, ADP and AMP in Postharvest Wax Apple (Syzygium Samarangense Merr.et Perry.) Fruits
HPLC (High Performance Liquid Chromatography) method was used for measuring the adenylic acid,energy charge and its influence on the cottony softening of wax apple(Black pearl) after harvest.The chromatographic separation was achieved on an Waters Atlantis T3 column (5.0 μm,4.6 mm×250 mm),which using a mobile phase composed of phosphate buffer (20 mmol/L KH2PO4:20 mmol/L Na2HPO4,pH 7.0)at a flow rate of 1.2 mL/min.Column temperature was set as 30 ℃ and the indirect UV detection was operated at 254 nm.Results indicated that the calibration curves displayed good linearity over a concentration range of 6.25~200 μg/mL.The content of ATP,ADP and AMP remained stable in wax apple sample extracted by boiling water,and the addition recovery for ATP was 93.71%(RSD=1.74%),ADP 97.28%(RSD=3.42%),AMP 98.17%(RSD=3.12%) separately.This method can be applied to determine the adenylic acid in wax apple,which was accurate,sensitive and convenient.
