响应面法优化超声辅助提取木耳黑色素

以木耳粉末为材料,通过单因素试验筛选出氢氧化钠浓度、超声功率、超声时间和料液比4个因素,再对后3个单因素进行Box-Behnken设计,采用响应面法优化超声波辅助提取木耳黑色素的工艺条件.试验结果表明,木耳黑色素的最佳提取工艺条件是:氢氧化钠浓度0.5 mol/L,超声功率348 W,超声时间9 min,料液比1:21g/mL.此条件下木耳黑色素吸光度为0.787,比传统溶剂法提取黑色素的提取率高26.94%.超声波辅助提取木耳黑色素的方法准确,节约溶剂且提取率高.
Optimization of Ultrasound-assisted Extraction of Melanin by Auricularia auricula Response Surface Method
melanin is a biologically active substance in Auricularia auricula with anti-aging and regulating immune function.The thesis tells new process conditions of ultrasonic assisted extraction of melanin,providing the technical basis for further development and utilization of Auricularia auricula resources.By Auricularia auricula powder materials,and by single-factor test filtering out sodium hydroxide concentration,ultrasonic power,ultrasonic time and solid-liquid ratio of 4 factors,then after 3 Box-Behnken single factor design using response surface methodology to optimize conditions of ultrasonic assisted extraction of melanin.Experimental results show that the optimum extraction technological conditions of melanin is the sodium hydroxide concentration 0.5 mol· L-1,the ultrasonic power 348W,ultrasonic time 9min and solidliquid ratio 1∶21 g/mL.Melanin absorbance of Auricularia auricula in this condition was 0.787,Extraction rate of the ultrasonic assisted extraction of melanin from A uricularia auricula increased by 26.94% compared with solvent extraction.The method of ultrasonic-assisted extraction of melanin is accurate,solvent-saving and efficient.
