黑李提取物对结肠癌Caco-2细胞增殖和调亡的影响

采用80%丙酮浸提法制备黑李粗提物.通过MTT法测定黑李提取物对Caco-2细胞增殖的抑制效果,Annexin V-FITC/PI双染色法、PI染色法、酶联免疫分析法分别检测细胞凋亡、细胞周期以及凋亡蛋白相对表达量变化,探究黑李提取物对Caco-2细胞凋亡信号转导通路的影响.与空白对照组相比,不同浓度的黑李提取物(0.2~2.0 mg/mL)对Caco-2细胞增殖均有明显的抑制作用,随浓度的增大,Caco-2细胞凋亡率明显升高,呈现显著的剂量-效应关系.提取物处理细胞72h后,使细胞周期G1期和G2延长,CytC、caspase-9、caspase-3等凋亡蛋白的相对表达量明显上升.提示黑李提取物通过阻滞细胞周期,诱导线粒体通路介导的细胞凋亡.本结果为将黑李开发为抗癌功能性食品提供了科学依据.
Abstract
To explore the anti-cancer activity and apoptosis of black plum against human colon cell Caco-2.Method: black plum extract was acquired by 80% acetone extraction, and inhibition effect on cell proliferation was determined by MTT method.In addition, to determinate cell apoptosis, cell cycle and expression of apoptosis protein, Annexin V-FITC/PI double staining assay, PI staining assay and ELISA were applied respectively.Result: Compared with control group, black plum extract had a significant inhibition on Caco-2 cell proliferation at different concentrations(0.2~2.0 mg/mL), and a dose-effect relationship can be observed.After treated with black plum extract for 72 h, cell cycle G1 and G2 periods were prolonged.The relative expressions of CytC, caspase-9, caspase-3 related to apoptosis showed obvious increases which proved that black plum extract can induce apoptosis via mitochondrial pathway.Conclusion: the result provides a scientific and theoretical basis to the development of anti-cancer functional food.
