丹酚酸B多重护肤功效的综合评价

目的:本文利用非酶糖基化反应模型、人永生化角质形成(HaCaT)细胞光老化模型、人胚胎皮肤成纤维细胞(ESF)内的胶原蛋白含量测定、酪氨酸酶抑制实验和细胞酪氨酸酶抑制实验对丹酚酸B(Sal B)的多重护肤活性进行了系统评价.方法:通过非酶糖基化反应模型评价了Sal B 抗糖基化效果;HaCaT 细胞光老化模型评价了Sal B 抗皮肤衰老活性;天狼猩红-苦味酸染色法测定ESF 中胶原蛋白含量,评价了Sal B 紧致皮肤的活性;蘑菇酪氨酸酶抑制实验、小鼠皮肤黑色素瘤细胞(B16-F10)中酪氨酸酶活性抑制实验,评价了Sal B 的美白作用,结合分子对接,推断了Sal B 抑制酪氨酸酶的作用位点.结果:Sal B 能抑制晚期高级糖化终末产物(AGEs)生成,具有抗糖基化作用,也能增强紫外损伤的HaCaT 细胞活力,具有明确的抗皮肤细胞光老化作用,其作用机制与提高细胞中的超氧化物歧化酶(SOD)、谷胱甘肽(GSH)等抗氧化物活性和含量有关.Sal B 还能增加ESF 中胶原蛋白含量,从而发挥紧致皮肤的功效.Sal B 可通过氢键作用、疏水作用等方式与酪氨酸酶结合,抑制其活性,从而发挥良好的美白功效.结论:本文利用多种模型综合评价了Sal B 的多重护肤功效,以期为Sal B 在护肤产品中的开发利用提供理论依据.
Comprehensive Evaluation of the Multiple Skin Care Effects of Salvianolic Acid B
Objective:This article systematically evaluates the multiple skincare activities of salvianolic acid B(Sal B)using non enzymatic glycosylation reaction models,human biochemical keratinocyte(HaCaT)cell photoaging models,collagen content determination in human embryonic skin fibroblasts(ESF),tyrosinase inhibition experiments,and cell tyrosinase inhibition experiments.Methods:The anti-glycation activity of Sal B was evaluated by nonenzymatic glycation reaction and the anti-aging activity of Sal B was evaluated by HaCaT cell photoaging model.The collagen content in human embryonic skin fibroblasts(ESF)was determined by sirius red-picric acid staining method,which determined the skin tightening activity of Sal B.The skin whitening effect of Sal B was evaluated by mushroom tyrosinase inhibition experiment and tyrosinase activity inhibition experiment in B16-F10 mouse melanoma cells.Combined with molecular docking,the sites of Sal B interacting with tyrosinase were predicted.Results:These results showed that Sal B could significantly inhibit the formation of advanced glycation end products(AGEs).It could also significantly enhance the viability of photoaging HaCaT cells by increasing the activity and content of endogenous antioxidants,including SOD and GSH.Sal B can also increase the content of collagen in ESF cells,thus exerting its skin tightening effect.Sal B can bind to tyrosinase through hydrogen bonds and hydrophobic interactions,significantly inhibiting its activity,thereby exerting a good skin whitening effect.Conclusion:In this paper,a variety of models were applied to evaluate the multiple skin care effects of Sal B,therefore providing a theoretical basis for the development and utilization of Sal B in skin care products.
