PI3K/Akt/mTOR Signaling Pathway and Its Role in Autophagy and Tumor
Autophagy is a self-digesting mechanism that characteristics of occurring double-membrane vesicles parceled long-lived proteins and damaged organelles and plays an essential role in maintaining cell homeostasis,development,tumorigenesis and infection.Recently,many studies have shown that autophagy acts as a “double-edged sword”.During tumor development and in cancer therapy,autophagy has paradoxically been reported to play roles in promoting both cell survival and cell death.PI3K/Akt/ mTOR pathway is a central regulating mechanism which is made of phosphoinositide 3-kinase (PI3K),protein kinase B(Akt) and mammalian target of rapamycin (mTOR),contributing to tumor growth and survival and inhibiting autophagy.This article gives an overview of PI3K/Akt/mTOR pathway and its links to autophagy and tumor.
