Simultaneous Determination of Lycopene,Lutein and Carotene in Tomato Products by Ultra Performance Liquid Chromatography
[Objective]The experiment established a rapid detection method for carotene in food using online solid-phase extraction and high-performance liquid chromatography technology with two pumps and one valve,which minimized the loss of carotene during the complex sample preparation.[Method]The solid-phase extraction column PLRP-S(2.1 mm×12.5 mm)and the liquid chromatography column PAH(2.1 mm×100 mm,1.8 μm)were used in this method.The mobile phase for online solid-phase extraction was water and methanol,and for liquid chromatography separation was methanol and dichloromethane.In this methodthe detection wavelength of 450 nm was applied.[Result]The regression equation of the standard curve maintained good linearity when the concentration ranging from 0.10 to 2.00 μg/mL.Also,both the limit of detection and the limit of quantification met requirements,and the recovery rate of α-carotene was between 92.0%and 105.4%,the recovery rate of β-carotene was between 91.1%and 107.2%.[Conclusion]This method utilized an online solid-phase extraction system with two pumps and one valve to efficiently and quickly test the content of carotene in food.The advantages are high detection efficiency and better extraction efficiency during sample preparation,so that the method can meet current detection demands.
