Prevention of High-fat Diet Induced Obesity and Dyslipidemia in Mice by Fucoidan
[Objective]To study the effect of fucoidan on obesity and lipid metabolism disorder induced by high-fat diet in mice.[Method]Mice were randomly divided into 3 groups including control group,high-fat diet group and fucoidan[100 mg/(kg·d)]treatment group, with 6 mice in each group.Mice in control group were fed basal diet, mice in other groups were fed high-fat diet, the mice in control group and high-fat diet group were given normal saline, the mice in fucoidan treatment group were given 100 mg/(kg·d)fucoidan,once a day by gavage, for 42 d continuously. The body weight and food intake of the animals were recorded during administration. At the end of administration, the fat-to-body ratio of mice was calculated, and the lipid levels[total triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C)and free fatty acids(FFA)]were measured in each group.[Result]The treatment of fucoidan could effectively reduce the weight gain(P<0.01)and fat-to-body ratio(P<0.05)of mice induced by high-fat diet. There was no significant effect on the food intake of mice(P>0.05),but the energy intake of mice in high-fat diet group was significantly higher than that in control group(P<0.01),while the food intake and energy intake of mice in Fucoidan treatment group were slightly lower than those in high-fat diet group, there was no significant statistical significance between the two groups(P>0.05).The serum TG,TC,LDL-C and FFA levels of mice in the high fat diet group showed different degrees of decline, while the HDL-C level was significantly increased.[Conclusion]Fucoidan polysaccharide has a strong preventive effect on obesity induced by high-fat diet in mice, and can also prevent the disorder of lipid metabolism caused by high-fat diet.
