Optimization of Ultrasonically Assisted Elderberry Polysaccharide Extraction Process and Its Structural Characteristics Analysis
[Objective]To optimize the optimal extraction process of ultrasonically assisted extraction of elderberry polysaccharides,and preliminarily study the structural characteristics of elderberry polysaccharides.[Method]Taking elderberry fruit(variety,Qingzhou No.15)as raw material,material-to-liquid ratio,ultrasonic temperature,ultrasonic time,and ultrasonic power as single factors,and designing the response surface on a single-factor basis to optimize the ultrasonic auxiliary extraction process,ultraviolet,infrared spectroscopy and high performance liquid chromatography were used for preliminary structural identification of butter polysaccharides.[Result]The optimal extraction process of elderberry polysaccharides was feed-to-liquid ratio 1∶20(g/mL),ultrasonic temperature 75 ° C,ultrasonic power 500 W,ultrasonic treatment for 45 min,and the yield of elderberry fruit polysaccharide up to 3.17%.Elderberry polysac-charide(EFP)is a α-glycoside bonded pyran-glycoside ring skeleton,and its monosaccharide composition is mainly glucose,arabinose,fucose,galactose,galactolic acid and mannose,and its ratio is 25.10∶23.20∶19.80∶12.60∶12.00∶6.40,and the molecular weight is 4.100×105 Da.[Conclusion]This study provides a theoretical basis for the development and utilization of elderberry and the structural characterization of elderberry polysaccharides.
