Rapid Analysis of Chemical Constituents of Rhodiola rosea Leaves by UPLC-Q-TOF-MS
[Objective]To determine the main phenolic,tannic and flavonoid components in Rhodiola rosea leaves by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS).[Method]80%methanol extract was separated by Waters BEH C18(2.1×100 mm,1.7 μm)with 0.1%formic acid water(A)-ethyl acid(B)as mobile phase gradient elution.The flow rate was 0.3 mL/min and the detection wavelength was 280 nm.Data were collected in negative ion mode.According to the molecular ion peaks and secondary fragment ions provided by high resolution mass spectrometry,and compared with literature or reference materials,the possible structure of the compound was determined.[Result]Totally 54 chemical components were identified from Rhodiola rosea leaves,including 11 phenolic acids,25 tannins and 18 flavonoids.This was the first systematic study of functional components in Rhodiola rosea leaves.[Conclusion]This method can provide scientific basis for the chemical constituents of Rhodiola rosea leaves cracking law and pharmacodynamic substance basis.
