A novel variant O allele of the ABO blood group gene associated with weak B antigen expression
Objective The aim of this study was to investigate the serology and gene sequence characteristics of the sample that discrepant results in forward/reverse ABO blood typing.Methods A blood donor with discrepant results in forward/reverse ABO phenotype was further investigated the complete ABO gene status by sequence based typing (SBT) and the haplotypes were sequenced by clone sequencing.Results The serological results of the sample suggested a Bweak type.Direct and clone sequencing analysis showed that the genotype was O01/O05,contained IVS6-28 A>G when compared with O01.Conclusion Apparent O alleles may cause weak B expression in red blood cells.Potential mechanisms for this condition requires further investigation.
