摘要

目的:优选女贞子多糖的最佳提取工艺并测定其单糖组成及摩尔比.方法:采用正交试验法,以多糖提取率为评价指标,对女贞子多糖的提取因素进行研究,用苯酚-硫酸比色法对女贞子多糖进行含量测定.多糖经硫酸水解后,水解产物利用高效液相色谱(HPLC)分析其单糖组成及摩尔比.结果:女贞子多糖的最佳提取工艺为10倍量水,回流提取3次,3 h/次;该多糖由鼠李糖、阿拉伯糖、甘露糖和葡萄糖4种单糖组成,其摩尔比为3.71:3.21:1.00:16.06.结论:该方法简便、准确,可用于女贞子多糖的单糖组成测定及质量控制.

Abstract

Objective; To optimize the extraction process of polysaccharide from Ligustrum lucidum Ait. (PLLA) and analyze monosaccharide compositionof PLLA. Method: With the extraction efficiency of polysaccharide as the index, an orthogonal design test was adopted to optimize the extraction process. The phenol-sulphuric acid colorimetric method was used to determine the content of PLLA. The PLLA was hydrolyzed by sulfuric acid, the hydrolyzate was assayed by HPLC. Result;The best extraction process of PLLA was added 10 times amount of water for 3 times and 3 hours for each time at the temperature of 100 ℃. PLLA composed of rhamnose, arabinose, mannose, glucose and their molratio is 3. 71: 3. 21: 1. 00: 16. 06. Conclusion; The method is simple, quick and accurate. This method could be applied for the analysis of monosaccharide composition and quality control in PLLA.
