Simultaneous Determination of Four Constituents in Geum aleppicum by RP-HPLC
Objective:To establish a method for simultaneously determining caffeic acid,quercetin-3-O-glucoside,kaempferol-3-O-glucoside and kaempferol in Geum aleppicum by RP-HPLC.Method:The quantitative analysis of G.aleppicum was carried out on Agilent C18 column (4.6 mm ×250 mm,5 μm),mobile phase was acetonitrile-0.5% formic acid with gradient elution,flow rate was 0.8 mL · min-1,columntemperature was maintained at 25 ℃,and detection wavelengrt was 340 nm.Result:The linear relationships of caffeic acid,quercetin-3-O-glucoside,kaempferol-3-O-glucoside and kaempferol fell within the ranges of 0.045-1.130,0.095-2.370,0.105-2.635,0.041-1.030 μg,respectively.Their average recoveries (n =6) were 99.0%,98.5%,97.7%,100.7% with the relative standard deviations (RSDs) of 1.0%,0.9%,1.0%,1.6%,respectively.Conclusion:This method is simple,accurate,sensitive,which can be used for quality control of G.aleppicum.
